چکیده
Abstract Study question Could the cell-free RNAs (cfRNAs) in seminal plasma be associated with male infertility and used as clinical biomarkers? Summary answer CfRNAs show different patterns men could serve diagnostic predictive factors infertility. What is known already Seminal a biological fluid containing large amounts of tissue-specific components secreted by organs reproductive system. Thus, has great potential sample for biomarker discovery noninvasive diagnostics disorders. Several metabolites, metal ions, proteins nucleotides are semen quality azoospermia forms subtypes. However, comprehensive transcriptome cfRNAs their value not clear. design, size, duration We applied previously developed library method polyadenylation ligation-mediated sequencing (PALM-Seq) to capture sequence from adult men, including 129 patients asthenozoospermia, 101 oligozoospermia, 102 (obstructive nonobstructive azoospermia) 107 healthy individuals. Participants/materials, setting, methods performed PALM-Seq on recruited individuals comprehensively profiled miRNA, tRNA, piRNA, lncRNA mRNA. In addition, we followed up outcome assisted technology (ART) during therapy. Main results role chance Hundreds miRNA tRNA detected all samples. Unlike other body biofluids, defined amount piRNA since highly expressed germ cells. Besides, showed group-specific pattern infertility, especially azoospermia. Differentially abundant mRNA oligozoospermia were mainly involved regulating mononuclear cell migration, thermogenesis development, respectively. The abundance was parameters quality, well sperm DNA fragmentation. At last, using collected omics data machine learning models, accurately classify subtypes successfully predict ART before treatment. Limitations, reasons caution only limited number this research more studies focused needed. Altered profile of seminal plasma microRNAs in the molecular diagnosis of male infertility.
BACKGROUND Although microRNAs (miRNAs) play essential roles in spermatogenesis, little is known about seminal plasma miRNAs in infertile men. We investigated the profile of seminal plasma miRNAs in infertile men to identify miRNAs that are altered in infertility; we then evaluated their diagnostic value. METHODS Seminal plasma samples were obtained from 289 infertile men and 168 age-matched f...
متن کاملSeminal plasma can be a predictive factor for male infertility.
Seminal plasma from ejaculates of 10 healthy, fertile volunteers and 63 infertile males was analysed for superoxide dismutase (SOD) and xanthine oxidase (XO) activities using a chemiluminometer. There was not statistically significant difference in the activity of either enzyme between control and infertile populations (113 +/- 74 IU/ml for SOD and 1.17 +/- 0.52 IU/ml for XO) in samples from no...
متن کاملRelationship of seminal plasma transferrin with seminal parameters in male infertility.
This study was undertaken to investigate whether there is relationship between seminal plasma transferrin and seminal parameters which included sperm count, motility and morphology. The study included 100 male subjects in the age group of 23-41 yrs including 7 proven fertility, 6 post-vasectomised and 87 subjects were of idiopathic infertility. Estimation of seminal plasma transferrin concentra...
متن کاملP-45: Cell Phones and Male Infertility
Background: Cell phones have become a vital part of everyday life. However, the health risks associated with their usage are often overlooked. Materials and Methods: MEDLINE, EMBASE, Cochrane Library and CINAHLPUBMED were searched for relevant trials published from respective database inception dates to Jun 2011. Results: Recently, evidence from several studies supports a growing claim that cel...
متن کاملReactive Oxygen Species and Antioxidant in Seminal Plasma and Their Impact on Male Fertility
Spermatozoa generate reactive oxygen species (ROS) in physiological amounts which play a role in sperm functions during sperm capacitation acrosome reaction (AR) and oocyte fusion. In addition damaged sperm are likely to be the source of ROS. The most important ROS produced by human sperm are hydrogen peroxide superoxide anion and hydroxyl radicals. Besides human seminal plasma and sperm posses...
